SNAP-8, also known as Acetyl Octapeptide-3, is a synthetic peptide developed by Lipotec (now part of Lubrizol Life Science) as a topical neuropeptide-based cosmetic active. It is structurally derived from SNAP-25, a synaptosomal-associated protein integral to neurotransmitter release at the neuromuscular junction.
SNAP-8 is an evolution of Argireline® (Acetyl Hexapeptide-8), with an extended peptide sequence designed to improve its inhibitory effects on muscle contraction, thereby reducing the appearance of expression lines and dynamic facial wrinkles. Unlike botulinum toxin (Botox), SNAP-8 is non-invasive, topically applied, and suitable for regular home use.
The SNARE (Soluble NSF Attachment Protein Receptor) complex facilitates vesicle docking and fusion with the presynaptic membrane, enabling acetylcholine release into the neuromuscular junction. SNAP-25 is a critical component of this complex, and by mimicking the N-terminal of SNAP-25, SNAP-8 competitively inhibits SNARE complex formation.
By destabilizing the SNARE complex:
SNAP-8 inhibits vesicle exocytosis
This leads to reduced acetylcholine release
Resulting in attenuated muscle contraction
As a result, repetitive facial muscle movements (which cause dynamic wrinkles) are visibly reduced, mimicking the effect of botulinum toxin in a reversible and localized fashion.
Clinical studies and manufacturer-sponsored trials report:
